在T细胞中，CyclosporinA与亲环蛋白(免疫亲和素)结合，形成亲环蛋白-CyclosporinA复合体，其结合并抑制钙调磷酸酶。CyclosporinA抑制钙调磷酸酶，IC50是7nM，随后阻断NF-AT核转运。CyclosporinA通过抑制MTP(线粒体通透性转换孔)的开放而影响线粒体，IC50是39nM。
